DRUG SENSITIVITY IN 3-D HISTOCULTURED TUMORS (Immuno-Oncology studies)

 

Histocultures are three-dimensional tissue cultures that maintain in-vivo-like phenotype in contrast to monolayer cultures. Histoculture tumor tissue contains cancer cells, stromal elements, immune cells interacting in their native environment.

Histocultured tumors can be tested for drug sensitivity to design individualized therapy for each patient or used as a powerful and cost-effective method in drug development [1]. Tumor response to mono or combination treatment could be studied using histoculture drug response assay (HDRA) [2].

Briefly, fresh tumor is sliced into thin pieces and incubated in the in the presence of tested compounds for the established period of time. An efficacy of anti-tumor compound is evaluated on the basis of tumor cell viability and/or any other parameter(s), such as induction of the specific gene expression, connected to the molecular mechanism(s) of compound’s anti-tumor activity.

Histoculture could be also a powerful tool for the development of pharmacodynamic markers.
